(96) Assay Card

General Information
Summary Studies using calcein assay and HeLa cells showed that at 10 uM, the increase in calcein leakage is very rapid.
Condition Calcein acetoxymethyl ester was added to incubated cancer cells. Cycloviolacin O2 was added at concentrations from 1 to 10 uM, and PBS was used as a control. Fluorescence of the entrapped calcein in each cell was measured with an Array Scan after 5, 15, 30 min, 1, 2, 3, and 4 h.
Result There was no leakage at 1 uM over 4 h. At 5 uM, there was a time-dependent increase in calcein leakage, which accelerated at higher concentrations. At 10 uM, the effect is very rapid.
