The cytotoxicity of cyclotides HB5 to HB9 were assessed against pancreatic cancer cells. Briefly, cells were seeded in 96-well flat-bottomed microtiter plates (2x10^3).After culturing for 12 h, cells were treated with cyclotides at 0.1, 0.5, 1, 4, 10 and 20 μM in 200 μL of media for 72 h. The medium was removed from each well and 100 μL of 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT; 0.5 mg/mL in PBS) was added in the absence of light; formazan crystals were produced over a 4h incubation period. To dissolve crystals, 150 μL of 0.04 N HCl in isopropanol was added to each well, and the optical density at 570 nm was measured on a spectrophotometer

IC50 of HB5 against cancer cell lines BxPC3, Capan2, MOH1 and PANC1 was 1.03 uM, 1.21uM, 1.32uM, 1.08uM.
IC50 of HB6 against cancer cell lines BxPC3, Capan2, MOH1 and PANC1 was 2.33 uM, 1.85 uM, 2.10 uM, 1.97 uM.
IC50 of HB7 against cancer cell lines BxPC3, Capan2, MOH1 and PANC1 was 0.68 uM, 0.45 uM, 0.33 uM, 0.36 uM.
IC50 of HB8 against cancer cell lines BxPC3, Capan2, MOH1 and PANC1 was 3.11 uM, 2.14 uM, 1.88 uM, 1.95 uM.
IC50 of HB9 against cancer cell lines BxPC3, Capan2, MOH1 and PANC1 was 1.54 uM, 1.32 uM, 1.14 uM, 2.01 uM. |
